چکیده:
هدف: در این پژوهش آثار تزریق دوطرفه آگونیست ها و آنتاگونیست های کانابینوئیدی در هیپوکامپ پشتی (CA1) بر حافظه موش های صحرایی حساس شده با آپومرفین بررسی شد. روش: روش اجتنابی مهاری (غیرفعال) با مدل دوطرفه برای بررسی حافظه در موش های صحرایی نژاد ویستار به کار گرفته شد و حافظه حیوان 24 ساعت بعد از آموزش مورد بررسی قرار گرفت. یافته ها: نتایج تزریق درون مغزی آگونیست گیرنده های CB1 و CB2، WIN55، 212-2 (µg/rat 5/0، 25/0) به صورت وابسته به مقدار به تخریب حافظه حیوانات در روز آزمون منجر شد. در حالی که تزریق درون مغزی آنتاگونیست اختصاصی CB1، AM251 (ng/rat 100، 50، 25) اثری روی حافظه نداشت. تزریق درون مغزی AM251، 2 دقیقه قبل از تزریق درون مغزی WIN55، 212-2 (µg/rat 25/0) اثری بر تخریب حافظه ناشی از WIN55، 212-2 (µg/rat 25/0) نداشت. هر چند که اثر تخریبی WIN55، 212-2 (µg/rat 25/0) به دنبال تزریق سه روزه آپومرفین (mg/kg، S.C 1، 5/0)، پنج روز قبل از تزریق WIM55، 212-2 کاملا از بین رفته و این اثر برگشتی حافظه در پی تزریق آنتاگونیست گیرنده D2 دوپامینی، سولپیراید (mg/kg، S.C 5/0، 25/0) 30 دقیقه قبل از تزریق آپومرفین مهار شد، در حالی که آنتاگونیست گیرنده D1، SCH23390 (mg/kg، S.C 1/0، 07/0، 02/0، 01/0) هیچ اثری بر پاسخ آپومرفین نداشت.
نتیجه گیری: هیپوکامپ پشتی نقش مهمی در فراموشی ناشی از کانابینوئیدها داشته و تزریق سه روزه آپومرفین ممکن است منجر به حساسیت گیرنده D2 دوپامینی شده و از این طریق در فراموشی ناشی از تحریک گیرنده CB1 اثر گذارد.
Objective: In present study، the effects of intra-dorsal hippocampal (intra-CA1) injection of cannabinoid receptor agents on memory retrieval have been investigated in 3-days apomorphine-treated rats.
Method: Passive avoidance task of memory has been used to examine the memory retrieval، 24 h after training. Apomorphine was injected subcutaneously (S.C.)، once daily for 3-days followed by 5 days free of the apomorphine before training. Results: Post-training intra-CA1 infusions of the non-selective CB1-CB2 receptor agonist، WIN55، 212-2 (0.1، 0.25 and 0.5 µg/rat)، dose-dependently shortened the step-through latency، suggesting impaired memory retrieval، whereas post-training intra-CA1 micro-injections of the selective CB1 receptor antagonist، AM251 (25، 50 and 100 ng/rat) did not affect memory retrieval. Intra-CA1 infusions of AM251 and WIN55، 212-2، two min apart، did not modify the WIN55، 212-2-induced reduction of step-through latency. However، the deleterious effect of WIN55، 212-2 (0.25 µg/rat) was completely abolished in rats previously given apomorphine (0.5 and 1 mg/kg/day، S.C.) for 3 days. This reversal of WIN55، 212-2-induced amnestic-like effect was counteracted by the dopamine D2 receptor antagonist، sulpiride (0.25، 0.5 and 1 mg/kg/day×3-days، S.C.)، administered 30 min before each injection of apomorphine (0.5 mg/kg/day×3-days، S.C.). The D1 receptor antagonist، SCH 23390 (0.01، 0.02، 0.07 and 0.1 mg/kg/day×3-days، S.C.)، was ineffective in this respect.
