Abstract:
زمینه و هدف: موسسه ملی بهداشت (NIH) اصطلاح بیماری قلبی- دیابتی را بهعنوان بیماری قلبی خاص در بیماران دیابتی تعریف کرده است که هایپرتروفی پاتولوژیک بطن چپ را هم شامل میشود؛ با این وجود فعالیت بدنی در بیماران دیابتی میتواند ساختار و عملکرد میوکارد را تحت تاثیر قرار دهد. با این حال، تنظیم مولکولی رشد فیزیولوژیکی قلب به عنوان یک پروتکل مکمل درمانی در مقابله با هایپرتروفی پاتولوژیک، کمتر درک شدهاست. از این رو در تحقیق حاضر اثر 8 هفته تمرین تناوبی با شدت بالا بر بیان ژن فسفاتیدیل اینوزیتول کیناز-3 (PI3K)، مقاومت به انسولین، گلوکز و انسولین سرمی در رتهای نر ویستار مبتلا به دیابت نوع 2 بررسی گردید. روش تحقیق: بیست سر رت نر نژاد ویستار با سن 10 هفته، در دامنه وزنی20±220 گرم به طور تصادفی به دو گروه شامل گروه تمرینات تناوبی با شدت بالا (HIIT) و گروه کنترل تقسیم شدند. پس از آشناسازی، گروه تمرینی در یک پروتکل ورزشی 8 هفتهای، 5 جلسه در هفته، به مدت 30 دقیقه در هر جلسه شرکت داده شدند. برای بررسی بیان ژن PI3K از روش RT-PCR، مقاومت انسولین روش ارزیابی مدل هموستازیHOMA –IR و سطوح سرمی گلوکز و انسولین از روش الایزا استفاده گردید. داده با آزمون t برای گروههای مستقل در سطح معنی داری 0/05>p مورد بررسی قرار گرفتند. یافتهها: در گروه HIIT افزایش معنیدار بیـان ژن PI3K نسبـت به گروه کنترل دیابتی تایید شد (0/003>p). همچنین تمرین باعث کاهش معنی دار مقاومت به انسولین (0/0001>p) و گلوکـز سـرم (0/0001>p) گردید؛ در حـالی کـه انسولیـن سرمی همگـام با این کاهش، افزایش معنی داری داشت (0/04p<). به علاوه، وزن قلب نیز افزایش معنی داری در گروه تمرینی پیدا کرد (0/0001p<). نتیجهگیری: تمرین HIIT با افزایش بیان ژن PI3K، مسیر مولکولی هایپرتروفی فیزیولوژیک را تحریک کرده و به عنوان یک درمان مکمل، در جلوگیری از هایپرتروفی پاتولوژیک بطن چپ نقش دارد.
Background and Aim: The term of diabetic heart disease has veen defined as the term of diabetic heart disease in the presence of a heart problem by the institute of National Health, particularly in some diabetic patient which pathologic left ventricular hypertrophy. However, physical activity can be affect structurally and functionally the myocardium in diabetic patients. Nevertheless; the molecular regulation of physiological heart growth has not been studied well as a supplement therapy protocol against pathologic hypertrophy. Therefore, the present study investigated the effect of 8 weeks of high intensity interval training (HIIT) on phosphatidylinositol kinase 3 (PI3K) gene expression, insulin resistance, heart weight, glucose and serum insulin in male Wistar rats with type 2 diabetes. Materials and Methods: Twenty male Wistar rats, 10 weeks old, weight 120±20gr were randomly divided into two groups including HIIT and control groups. After familiarization, the training group was participated in an 8-week of training protocol, 5 sessions per week, for 30 minutes per session. The gene expression of PI3K, insulin resistance, serum glucose and insulin levels were measured by the RT-PCR, HOMA -IR hemostasis, and ELISA methods respectively. The data were analyzed by the t-test for independent groups at pResults: The result indicated that in the HIIT group, PI3K expression was significantly higher than control group (pConclusion: HIIT exercise increases the expression of PI3K gene, stimulates the physiological molecular pathway of hypertrophy and could plays a role in preventing left ventricular hypertrophy as a complementary treatment.
