چکیده:
حضور هیستامین در مواد غذایی نه تنها میتواند موجب مسمومیت مصرف کنندگان شود، بلکه یکی از شاخصهای مهم فساد باکتریایی محسوب میشود. بنابراین شناسایی و اندازهگیری مقدار این آمین بیوژن از اهمیت بسزایی برخوردار است. در این تحقیق، برای اندازهگیری مقدار هیستامین تولیدی در محیط کشت TSB توسط باکتریهای مولد هیستامین، استافیلوکوکوس اپیدرمیدیس TYH1 بهعنوان سوش استاندارد مولد هیستامین و دو سوش استافیلوکوکوس کاپیتیس و استافیلوکوکوس کارنوسوس جدا شده از شیر گوسفند و دارای ژن مولد هیستامین، هیستیدین دکربوکسیلاز، از دو روش کروماتوگرافی مایع با عملکرد بالا (HPLC) بهعنوان روش مرجع و الکتروفورز موئینهای با آشکارساز جذبی (CZE) استفاده شد و یافتههای بهدست آمده از دو روش با هم مقایسه شدند. زمان مهاجرت در روش CZE و زمان بازداری در روش HPLC برای هیستامین بهترتیب 4/ 5 و 0/ 12 دقیقه و منحنی استاندارد در محدوده µg/ml 200- 6/25 بهصورت خطی با معادله رگرسیون y=0.000004x (r2=0.999) و y=0.000004x (r2=0.999) بهدست آمد. مقدار هیستامین تولید شده توسط سه سوش باکتری و اندازهگیری شده با دو روش مذکور تفاوت آماری معنیداری نداشت (P=0.39) و ضریب همبستگی بین یافتههای بهدست آمده از دو روش 99/ 0 بود. با توجه به یافتههای مشابه دو روش، روش CZE بهدلیل عدم نیاز به آمادهسازی نمونه، سادگی، حساسیت و کم هزینه بودن بهعنوان روشی مناسب برای اندازهگیری مقدار هیستامین در محیطهای کشت میکروبی پیشنهاد میگردد.
The presence of histamine in foods, as a chemical indication of food spoilage, can also cause food poisoning. Therefore, monitoring of histamine levels in food and food products is important. This paper has focused on the comparing of a fast and selective separation method, capillary zone electrophoresis (CZE), with HPLC, for the determination of histamine in TSB, inoculated with histamine producing strains including histamine forming gene, histidine decarboxylase-hdc, Staphylococcus epidermidis TYH1, isolated from fish miso in Japan, Staphylococcus capatis and staphylococcus carnosus, isolated from milk of sheep in Iran. The migration time of histamine in the proposed method, CZE and retention time of HPLC, were 5.4 and 12 min, respectively. The calibration graph was linear in a range of 6.25-200 μg mL-1 in CZE and HPLC method with same regression equation, y=0.000004x, and both methods showed a good agreement (r2=0.999). Determination of histamine produced by these three strains indicated no significant differences by both methods (P=0.39). The results show that the CZE is suitable for the determination of histamine in bacterial culture, as to perform the method, there is no need for initial preparation, simplicity, sensitivity and low cost.
