چکیده:
هدف: عدم تعادل در بیان PGC-1α و تولید مایوکاینها باعث کاهش مصرف انرژی، افزایش آدیپوسیتها و بروز اختلالات متابولیکی میشود، اما فعالیت بدنی میتواند از طریق PGC-1α، تولید مایوکینها را تنظیم کند. هدف از پژوهش حاضر بررسی اثر هشت هفته دویدن تداومی و تناوبی بر بیان ژن PGC-1α عضلانی و غلظت سرمی آیریزین در موشهای صحرایی نر مبتلا به سندرم متابولیک بود. روش ها: تعداد 32 سر موش صحرایی نر نژاد ویستار 6 هفته ای با وزن 180-150 گرم، به طور تصادفی به دو گروه رژیم غذایی استاندارد (8n=) و رژیم غذایی با چربی بالا (24 (n= تقسیم شدند و به مدت 12 هفته تحت رژیم غذایی قبل از ورزش قرار گرفتند. سپس 24موش صحرایی گروه رژیم غذایی با چربی بالا به طور تصادفی به سه گروه مساوی کنترل سندرم متابولیک (Ctr+MS)، تمرین تداومی (CT-MS) و تمرین تناوبی (IT-MS) تقسیم گردیدند. تمرینات تداومی و تناوبی به مدت هشت هفته، به ترتیب باشدت معادل 75-65 و 100-80 درصد سرعت بیشینه روی نوارگردان مخصوص موشهای صحرایی اجرا شد. جهت انـدازه گیـری آیریـزین سـرم از روش ELISA و بیان ژن PGC-1α از RT-PCR استفاده شد. آزمونهای آماری تحلیل واریانس یک راهه، آزمون تعقیبی توکی و همبستگی پیرسون در سطح معنا داری05/0 p≤ برای تجزیه و تحلیل دادهها به کار گرفته شد. نتایج: پس از هشت هفته تمرین تناوبی و تداومی بیان PGC-1α (به ترتیب01 /0=P و 001/ 0=P) و سطح سرمی آیریزین (به ترتیب01/ 0=P و 001/ 0=P) نسبت به گروه کنترل سندرم متابولیک افزایش یافت، این افزایش در گروه تمرین تناوبی نسبت به تمرین تداومی بالاتر بود (001/0=P). همچنین بیان عضلانی PGC-1α همسبتگی مثبت با سطح سرمی آیریزین موشهای صحرایی داشت (8/ 0r=، 01/0 =P). نتیجهگیری: به نظر میرسد استفاده از تمرینـات تناوبی میتواند به اندازه تمرینات تداومی و حتی بیشتر باعث افزایش بیان ژن PGC-1α و آیریزین و احتمالا بهبود اختلالات متابولیکی در افراد مبتلا به سندروم متابولیک گردد.
Purpose: An imbalance in the production of PGC-1α and myokines impairs the energy expenditure, increases adipocyte and develops metabolic pathologies, but physical exercise is able to regulate the secretion of myokines through PGC-1α. The aim of this study was to evaluate the effect of eight weeks of high-intensity interval training vs. continuous training on serum Irisin levels and expression of skeletal muscle PGC-1α gene in male rats with metabolic syndromeMethods: 32 Wistar male rats, with six weeks’ age and weight 150-180 gr, randomly divided into two groups including standard diet (n=8) and high fat diet (n=24). They were then placed on a pre-exercise diet for 12 weeks. 24 rats from high fat diet group were divided randomly into two groups including metabolic syndrome control (Ctr+MS), continuous running (CT-MS) and Interval running (IT-MS). Continuous and interval training for eight weeks were performed on 65-75 and 80-100% of the maximum speed on the treadmill for mice, respectively. The ELISA method for measuring serum Irisin levels and real-time PCR method for expression of PGC-1α gene were used. The data were analyzed by one-way analysis of variance (ANOVA), Tukey's post-hoc test and Pearson correlation at P Results: After eight weeks of interval and continuous training, PGC-1α expression (P = 0.01 and P= 0.001) and serum levels of Irisin (P = 0.01 and P= 0.001) increased compared to the metabolic syndrome control group, and this increase was more evident in the interval group than the continuous group (P = 0.001). Muscle expression of PGC-1α was also positively correlated with serum Irisin levels in mice (P = 0.001, r = 0.8).Conclusion: It seems that the use of interval exercises can increase the expression of PGC-1α gene and Irisin as much as continuous exercise and possibly improve metabolic disorders in people with metabolic syndrome.
