چکیده:
سابقه و هدف: نانوذرات نقره کاربرد وسیعی در علوم پزشکی، دارویی و بهداشتی دارند. معمولا در روشهای فیزیکی و شیمیایی ساخت نانوذرات، مشکلاتی از جمله آلودگی محیط زیست، هزینه بر بودن و پیچیده بودن فرآیند سنتز وجود دارد. تولید زیستی نانوذرات نسبت به این روشها، ساده، کم هزینه و عاری از خطرات زیست محیطی می باشد. هدف از پژوهش حاضر تولید زیستی نانوذرات نقره با استفاده از باکتری باسیلوس می باشد.
مواد و روشها: باکتری های جنس باسیلوس با استفاده از مشاهدات مورفولوژیکی و آزمایشات اولیه تشخیصی بیوشیمیایی از خاکهای منطقه آینالی و معادن مس سونگون و مزرعه شهر اهر جداسازی شدند. سپس در محلول نیترات نقره 1 و 5/3 میلی مولار با pH برابر با 7 به مدت 72-48 معلق شدند. پس از تغییر رنگ محلول واکنش، بیومس باکتری با استفاده از اسپکتروفوتومتری UV-vis و تصویربرداری با میکروسکوپ الکترونی TEM مورد بررسی قرار گرفت. آزمایشات مولکولی، جهت تایید جنس باکتری مورد نظر، انجام گردید.
یافته ها: از 40 باکتری باسیلوس جداسازی شده از نمونه های خاک، تنها یک گونه جدا شده از معدن مس سونگون قادر به تولید نانوذرات نقره بود. نتایج به دست آمده از طریق تغییر رنگ محلول واکنش، اسپکتروفتومتری UV-vis و تصاویر میکروسکوپ الکترونیTEM نشان داد که باکتری باسیلوس مورد مطالعه، قادر به احیاء یون نقره به نانوذرات نقره به دو صورت داخل سلولی و خارج سلولی است. نانوذرات نقره تولیدی کروی شکل بوده، بیشتر بر روی دیواره خارجی سلول باکتری تجمع کرده و اندازه آنها در گستره 50-20 نانومتر بود.
نتیجه گیری: نتایج نشان داد باکتری باسیلوس جداسازی شده از معدن مس سونگون اهر قادر به تولید نانوذرات نقره می باشد.
Background and Aim: Silver nanoparticles have many applications in medicine، pharmaceutical and health sciences. There are some problems in physical and chemical methods of nanoparticle production including high cost، environmental pollution and complexity of these processes، while the biological production methods are economical and eco-friendly. The aim of this study was biosynthesis of silver nanoparticles by Bacillus bacteria.
Materials and Methods: The gender of Bacillus bacteria was isolated from Ainaly territory soil and copper mines of Ahar city (Sungun and Mazraeh) using morphological observation and some preliminary biochemical tests. The isolated bacteria were suspended in 50 ml of 3.5 mM aqueous AgNo3 solution at pH 7 for 48-72 h. After color changing observation، the solutions were centrifuged to prepare biomass for analyses of UV-vis spectrophotometer and TEM. The molecular test was performed to confirm the gender of the bacteria.
Results: After isolation of 40 kinds of Bacillus bacteria category، just the one from Sungun Copper Mine was found to be able to produce silver nanoparticles. The colour changing of aqueous medium and the result of UV–vis spectrophotometer demonstrated the formation of silver nanoparticles. TEM micrographs indicated spherical silver nanoparticles with the size range of 20-50 nm. The molecular test demonstrated the genus of bacteria as Bacillus.
Conclusion: The results showed that the isolated species of Bacillus is able to produce sliver nanoparticles.
