چکیده:
تنظیم نامناسب در پویایی میتوکندری با اختلال در تنظیم متابولیسم همراه است و به بیماریهای متابولیکی منجر میشود. هدف پژوهش حاضر بررسی تاثیر تمرین هوازی بر بیان ژن Drp1، Cyp1a1 و بیان پروتئین CYP1A1 عضلة نعلی رتهای نر دیابتی است. در این مطالعة تجربی 24 سر رت نر ویستار بهطور تصادفی به 3 گروه دیابت (D)، گروه کنترل (Ct)، و گروه تمرین+دیابت (ED) تقسیم شدند. مدل دیابت در رتها با تزریق STZ القا شد. گروه ED به مدت 8 هفته، 5 جلسه در هفته پروتکل تمرین هوازی را اجرا کردند. پس از اتمام پروتکل، عضلة نعلی رتها استخراج، و از روش RT-PCR، برای سنجش بیان ژن Drp1 و Cyp1a1 و از روش الایزا برای سنجش بیان پروتئین CYP1A1 استفاده شد. میزان RNA با استفاده از فرمول محاسبه و سپس دادهها با استفاده از آزمون تحلیل واریانس تجزیهوتحلیل شد. نتایج تفاوت معناداری را بین گروهها در بیان ژن Drp1 (001/0P£) و Cyp1a1 (017/0P£)، بیان پروتئین CYP1A1 (028/0=P) و گلوکز (001/0P£) نشان داد. دیابت بیان ژن Drp1 و Cyp1a1 و بیان پروتئین CYP1A1 را در عضلة اسکلتی افزایش داد. این در حالی است که تمرین فقط بیان ژن Drp1 را کاهش داد (013/ 0=P). دیابت به افزایش گلوکز، بیان ژن Drp1 و Cyp1a1 و بیان پروتئین CYP1A1 در عضلة اسکلتی منجر میشود. اما تمرین، تنها از افزایش بیان ژن Drp1 پیشگیری کرد.
Improper regulation of mitochondrial dynamics is associated with dysregulation of metabolism and can lead to metabolic diseases. This study aimed to investigate the effect of aerobic training on gene expression of Drp1 and Cyp1a1 and protein expression of CYP1A1 in soleus muscle of male diabetic rats. In this experimental study, 24 male Wistar rats were randomly divided into 3 groups: diabetes (D), control (Ct), and exercise + diabetes (ED) group. The diabetic model was induced in rats by STZ injection. The ED group performed an aerobic training protocol five times a week for eight weeks. After the end of the protocol, rat soleus muscle was extracted, and gene expression of Drp1 and Cyp1a1 RT-PCR and protein expression of CYP1A1 were measured using ELIZA. RNA was calculated using the formula 2^(-ΔΔCT)and then the data were analyzed using ANOVA. The results showed a significant difference between groups in the gene expression of Drp1 (P0.001) and Cyp1a1 (P=0.017), protein expression of CYP1A1 (P=0.028), and glucose (P0.001). Diabetes increases the gene expression of Drp1 and Cyp1a1 and the protein expression of CYP1A1 in skeletal muscle. However, exercise could only reduce gene expression of Drp1 (P=0.013). Diabetes leads to increased glucose, gene expression of Drp1 and Cyp1a1, and protein expression of CYP1A1 in skeletal muscle. But the exercise could prevent only increase of gene expression of Drp1.However, exercise could only reduce Drp1 expression (P=0.013). Diabetes leads to increased glucose, gene expression of Drp1 and Cyp1a1, and protein expression of CYP1A1 in skeletal muscle.

فرایندهای مختلفی همجوشی و شکافت ١ریخت شناسی و عملکرد میتوکندری را تنظیم میکنند و نشان داده شده است که تمرین هوازی محتوی میتوکندری عضلۀ اسکلتی و آنزیم های اکسیداتیو را افزایش میدهد و با افزایش بیان پروتئین های درگیر در پیامرسانی انسولین ، به بهبود چشمگیر اکسیداسیون گلوکز و اسیدهای چرب منجر می شود (٧).
اما همانطورکه در نتایج پژوهش حاضر مشاهده میشود، ورزش اثری بر تغییرات ناشی از دیابت برCYPA١١ نداشته است که احتمالا یکی از دلایل افزایش CYPA١١ در رتهای دیابتی ناشی از اثر سمی STZ باشد که تمرین استقامتی نیز نتوانسته است تغییر معناداری در این متغیر ایجاد کند.
در کل نتایج پژوهش حاضر نشان داد که هر دو متغیر Drp١ و CYPA١١ تحت تأثیر بیماری دیابت قرار دارند و تمرین استقامتی به تنهایی فقط برDrp١ در عضلۀ اسکلتی تأثیرگذار است و برای تغییر در CYPA١١ نیاز به روشهای درمانی دیگر احساس می شود، که باید در تحقیقات آینده بررسی شود.
