چکیده:
مقدمه: سرب یکی از چهار فلزی است که بیشترین عوارض را بر سلامت انسان دارد. با آنکه هیچ نقش حیاتی برای آن شناخته نشده، میتواند در سیستمهای زیستی آنقدر تجمع یابد تا به حد سمی برسد. سرب از طرق مختلف وارد محیط زیست میشود. یکی از روشهای سنجش آلودگی فلزات سنگین در محیط، زیست حسگر سلولی است که سلول حاوی یک ژن گزارشگر تحت کنترل پروموتر حساس به یک عامل (نظیر فلزات سنگین) میباشد. یکی از اپرونهای مهم در مقاومت باکتری رالستونیا به فلزات سنگین، اپرون pbr است و هدف از این مطالعه طراحی زیست حسگر حاوی ناحیه پروموتری pbr به همراه ژن گزارشگر لوسیفراز میباشد و از باکتری E.coli سویه DH5α بهعنوان میزبان استفاده گردید. مواد و روشها: توالی پروموتری pbr به همراه ژن تنظیمی pbrR با سایز 634 نوکلئوتید سنتز شد و ژن گزارشگر لوسیفراز در پلاسمید pGL3، تحت این ناحیه پروموتری قرار گرفت سپس پلاسمید نوترکیب به باکتری E.coli سویه DH5α منتقل شد. باکتری حاوی پلاسمید نوترکیب قادر به تشخیص سرب در محیط میباشد و از زیست حسگر ساختهشده بهمنظور سنجش غلظتهای مختلف سرب در محیط کشت استفاده گردید. یافتهها: کمترین غلظت سرب که قادر است باعث بیان معنیدار ژن گزارشگر لوسیفراز شود، یک میکرومولار بود و حداکثر بیان ژن گزارشگر در غلظت 100 میکرومولار مشاهده گردید. به نظر میرسد غلظت بالاتر از 100 میکرومولار، بهواسطه اثرات سمی بر بقای حسگر باعث کم شدن بیان ژن گزارشگر گردیده است. نتیجهگیری: نتایج حاصل از این پژوهش نشان داد که این حسگر قابلیت سنجش غلظتهای یک تا 100 میکرومولار فلز سنگین سرب در محیطهای آبی را دارد.
Background: Lead is one of the four metals that has most compli-cation on human health. Although there has been no known vital ef-fect for it, lead can be collected in living systems to be toxic. Lead enters the environment by different approaches. One of the methods for heavy metals impurity detection in environment is cellular bio-sensor. Cellular biosensor is cell containing one reporting gene under the control of promoter that is sensitive to an element (such as heavy metals). One of the important operons in Ralstonia bacterium re-sistance to heavy metals is pbr. This research was done to design bio-sensor containing pbr promoter with Luciferase reporting gene in E.coli (DH5α) as host.
Materials and Methods: The pbr promoter sequence with pbrR as regulator gene in 634 nucleotide size was synthesized and Luciferase reporting gene in pGL3 plasmid was located under this promoter. Then the recombinant plasmid was transferred to E.coli (DH5α). These bacteria were able to detect the lead in the environment and were used to detect different concentrations of lead in medium.
Results: The least concentration of lead that can significantly ex-press Luciferase reporting gene was 1μM and the most concentration of it was 100μM. It seemed that the concentration above 100μM caused decrease in reporting gene expression through toxic effects on biosensor survival.
Conclusion: The results indicated that this biosensor can detect 1-100 μMol concentration of the heavy metal "Lead" in aqueous solu-tions.
