چکیده:
سابقه و هدف: با وجود پیشرفت های گسترده در زمینهی کشت سلول های بنیادی، هنوز انتخاب بهترین منبع سلولی کارآمد جهت تزریق، قابل تامل است. علاوه بر این، تا کنون از نشانگر کلسیمی برای بررسی سلول های عصبی حرکتی استفاده نشده است. روش هایی که تا کنون برای بررسی عملکرد سلولی مورد استفاده قرار گرفته همچون روش های الکتروفیزیولوژی (patch-clamp)، علاوه بر سخت بودن و وقت گیر بودن، در هر بررسی فقط یک سلول را مورد بررسی قرار می دهد. در این تحقیق، نشانگر کلسیمی و ترشح وزیکول های سیناپسی در سلول های عصبی- حرکتی مشتق از بافت چربی، با استفاده از رنگ آمیزی FM1-43 و نشانگر کلسیمی FLUO-2 مورد بررسی قرار گرفته است.
مواد و روش ها: از موش صحرایی نر بالغ، نژاد wistar با وزن 250-200 گرم استفاده شد. بافت چربی استخراج و سلول های بنیادی آن جداگردید. سپس، سلول های به دست آمده از لحاظ مارکرهای سلول های بنیادی مزانشیمی و مولتی پوتنسی، مورد بررسی قرار گرفتند. سلول های بنیادی مشتق از بافت چربی تحت تاثیر القا کننده های رتینوئیک اسید و SHH، به سلول های شبه عصبی حرکتی تمایز یافتند. بعد از تمایز، سلول های تولید شده با استفاده از تکنیک های ایمنوسیتوشیمی و RT-PCR مورد تایید قرار گرفتند. در مرحله ی نهایی، فانکشنال بودن سلول ها با توانایی برقراری ارتباط عصبی-عضلانی، ترشح وزیکول های سیناپسی و تبادلات کلسیمی، مورد ارزیابی قرار گرفت که برای این کار از روش های هم کشتی، رنگ آمیزی FM1-43 و نشانگر کلسیمی FLUO-2 استفاده گردید.
یافته ها: سلول های عصبی- حرکتی مشتق از بافت چربی، تمام مارکرهای مزانشیمی و اختصاصی چربی را بیان می کنند و توانایی تمایز به سایر رده های سلول های مزانشیمی شامل سلول استخوانی، غضروفی و چربی را دارند. این سلول ها، بعد از تمایز به سلول عصبی- حرکتی، مارکرهای islet-1، oligo-2 و HB9 را بیان می کنند. همچنین، دارای توانایی برقراری ارتباط عصبی- عضلانی، ترشح وزیکول های سیناپسی و تبادلات کلسیمی می باشند.
نتیجه گیری: نتایج بدست آمده نشان می دهد که سلول های عصبی- حرکتی مشتق از بافت چربی، توانایی برقراری ارتباط عصبی- عضلانی، ترشح وزیکول های سیناپسی و تبادلات کلسیمی را داشته و یک سلول فانکشنال می باشند.
Background and Aim: Cell replacement therapy has provided the basis for future clinical applications to treat neurodegenerative diseases، such as motoneuron diseases. Induced functional motoneurons are an option for replacing the lost motoneurons. Adipose derived stem cells (ADSCs) are an appropriate source of cells for autologous cell therapy with the ability of neural differentiation. In this study we use calcium ion imaging and synaptic vesicle release in order to indicate the functionality of motoneurons from adipose derived stem cells.
Materials and Methods: In the present study، ADSCs were induced to motoneuron-like cells (MNLCs) by SHH and retinoic acid (RA) as inducers. ADSCs markers such as CD90، CD 49d، CD106، CD45، were measured by immunocytochemistry analysis and their multipotency were evaluated by incubation of the ADSCs with adipogenic، chondrogenic، osteogenic induction media. In addition to RT-PCR، immunocytochemistry was utilized in order to approve the expression of islet-1، oligo-2 and HLXB9 in induced MNLCs from ADSCs. To identify the functional MNLCs، a co-culture preparation of MNLCs and myocytes، calcium ion imaging and synaptic vesicle release was used.
Results: ADSCs treated with a mixture of preinducer (B27، EGF، and bFGF) and inducers factors (SHH and RA) adopted a morphology similar to motoneuron cells. Immunocytochemical staining and RT-PCR approved the treated cells expressed the motoneuron markers islet-1، oligo-2 and HLXB9. The co-cultured with myocytes indicates the formation of neuromuscular connections between MNLCs and myocytes. After two week، MNLCs showed high HLXB9 expression، indicative of full differentiation. Also، the release rate of synaptic vesicles using FM1-43 in the induced MNLCs was approved. Moreover، calcium imaging with fluo-2 results approved that functional excitatory synaptic connections can influence the activity of MNLCs.
Conclusion: These results indicate ADSCs can be differentiated to a functional MNLCs phenotype and may be of benefit for treatment of motoneuron diseases.
