چکیده:
مقدمه: فارنسیفرول C یکی از ترکیبات بیولوژیکی فعال مستخرج از ریشه گیاهFerula szowitziana میباشد که دارای اثرات ضدسرطانی متعددی از جمله مهار تکثیر سلولی و فعالسازی آپوپتوز است. با این حال، این ترکیب گیاهی محلولیت و زیستماندگاری ضعیفی در محیطهای in vitro وin vivo دارد. نانوذرات دندروزومی قادر به بالا بردن پایداری ترکیبات هیدروفوب گیاهی از جمله کومارین و در نتیجه خواص ضدسرطانی آنها میباشند. هدف از مطالعه حاضر، افزایش محلولیت و سمیت فارنسیفرولC از طریق بهکارگیری نانوذرات دندروزوم و به دنبال آن ارزیابی اثرات سیتوتوکسیکی آن روی سلولهای سرطانی آدنوکارسینومای معده میباشد. مواد و روشها: سمیت سلولی ترکیب دندروزومی فارنسیفرول C در مقایسه با فرم آزاد آن روی سلولهای AGS مورد بررسی قرار گرفت. بدین منظور، سلولهای AGS در محدوده غلظتیµΜ 150-0 از فارنسیفرول C و در بازههای زمانی 24 و 48 ساعت تحت تأثیر قرار گرفتند و با استفاده از تست MTT میزان سمیت آنها بهصورت IC50 ارزیابی گردید. دادهها با استفاده از برنامه آماری رگرسیون و t-test تجزیه و تحلیل شدند. یافتهها: دادههای بهدستآمده نشان داد که بین دو متغیر غلظت فارنسیفرول C و درصد سلولهای زنده رابطه معکوس و معنیداری وجود دارد (0/001 ≥ P). همچنین، اثر کشندگی نوع دندروزومی فارنسیفرولC در مقایسه با فرم آزاد آن روی سلولهای سرطانی AGS بهمراتب بیشتر بوده و در دوزهای پایینتری باعث مرگ سلولها شده است (0/05 ≥ P). نتیجهگیری: نتایج اولیه بهدستآمده از این تحقیق نشان میدهد که فارنسیفرول C میتواند کاندیدای دارویی مناسب و بیخطری در جهت از بین بردن سلولهای سرطانی محسوب شود که نیازمند تأیید مولکولی سمیت آن در سطح سلولهای مختلف سرطانی و نرمال میباشد.
Background: Farnesiferol C, a sesquiterpene coumarin, is one of the biologically active compounds extracted from the roots of Ferula Szowitziana and shows different anti-cancer properties such as anti-proliferation and apoptosis induction. However, the solubility and bioavailability of this phytochemical is poor in vivo and in vitro. Denderosome nanoparticles could improve the solubility of Farne-siferol C and hence its anti-cancer properties. The aim of current study was based on improving the solubility of Farnesiferol C by tak-ing advantage of dendrosome nanoparticles; and subsequently evalu-ating its cytotoxic effect against gastric adenocarcinoma.
Materials and Methods: The cytotoxic effect of dendrosomal Far-nesiferol C was investigated on AGS cells by MTT assay. The cells were then incubated with fresh medium containing serial concentra-tions of Farnesiferol C (0 to 150 μM) in the form of dendrosomal and void Farnesiferol C for 24 and 48h. Afterward, the toxicity of this botanic compound and its IC50 were measured. Data were analyzed by regression & t-test.
Results: The data illustrate a reverse and significant relation be-tween dendrosomal Farnesiferol C and void Farnesiferol C variables (P<0.001). Also, cytotoxic effect of dnedorsomal Farnesiferol C on AGS cells was much more than free Farnesiferol C and affected in low concentrations (P<0.05).
Conclusion: The results showed that dendrosomal Farnesiferol C could be an appropriate and safe anti-cancer phytochemical that de-mands molecular evaluation of its anti-tumor effect on different can-cerous and normal cell lines.
