چکیده:
سابقه و هدف: تاکنون انواع مختلفی از غشاهای غیر قابل جذب به منظور بازسازی ساختار بافت پریودنتال آسیب دیده مورد استفاده قرار گرفته است. هدف از مطالعه تجربی حاضر، مقایسه ی انواع غشاهای غیر قابل جذب از نظر میزان و نحوه ی چسبندگی سلول های PDL (perio dental ligament) انسانی در محیط آزمایشگاهی می باشد. برای نیل به این هدف، میزان نرخ بقاء، واکنش آلکالین فسفاتاز و مورفولوژی سلول ها پس از مجاورت با غشاهای مورد نظر، بررسی شدند.
مواد و روش ها: سه غشای غیر قابل جذب GBR-200، TXT-200 و Imtec Ovid مورد مطالعه قرار گرفتند. سلول های فیبروبلاست جدا شده از PDL دندانهای پرمولر با دانسیته ی 104 cell/ml در محیط کشت بر روی غشاها با ابعاد 33 میلی متر در 15 چاهک برای 24 ساعت انکوبه گردید. سپس نمونه ها برای ارزیابی توسط میکروسکوپ نوری و الکترونی فیکس شدند. داده ها بوسیله ی آزمون آماری one-way ANOVA آنالیز گردیدند.
یافته ها: نتایج نشان داد که از 3 غشای مورد بررسی، غشاهای TXT-200 و GBR-200 بیشترین میزان چسبندگی را به لحاظ مورفولوژی داشته و کمترین چسبندگی مربوط به غشای Imtec Ovid بود که این تفاوت از نظر آماری معنی دار بود (02/0p=). همچنین، میزان چسبندگی سلول ها از نظر شاخص ALP (alkaline phosphatase) و MTT (3-(4،5-Dimethylthiazol-2-yl)-2،5-Diphenyltetrazolium Bromide)، بر روی غشاهای TXT-200 و GBR-200 بیشتر از گروه دیگر بود، اما این تفاوت از نظر آماری معنی دار نبود (به ترتیب 2/0p= و 08/0p=).
نتیجه گیری: به نظر می رسد غشاهای TXT-200 و GBR-200 نتایج بهتری را در تغییرات مورفولوژیک سلول های فیبروبلاست در محیط کشت نشان دادند. شاید علت معنی دار نبودن شاخص های ALP و MTT ، کم بودن تعداد سلول های چسبیده به غشاها باشد که در نتیجه امکان اندازه گیری دقیق این شاخص ها را میسر نمی ساخت.
Background and Aim: Different types of non-bioabsorbable membranes have been widely used for guided tissue regeneration (GTR) with its ultimate goal of regenerating lost periodontal structures. The purpose of this present in- vitro study is to evaluate the effects of various non-bioabsorbable membranes on the quality and quantity of the human periodontal ligament fibroblast cells attachment.
Materials and Methods: Three non-bioabsorbable membranes (TXT-200، GBR-200، IMTEC OVID] were studied. All membrane with the dimension of 3×3 mm were exposed to human periodontal ligament fibroblast from extracted premolars with cell density of 105 cell/ml in 15 wells، and then incubated for 24 hours. Moreover، samples were fixed and processed for light and scanning electron microscopy evaluation. Data were statistically analyzed using the one- way ANOVA test.
Results: The results showed three barriers were tested، in over times، TXT-200 and GBR-200 membranes had the highest rate of attachment and IMTEC OVID membrane had the lowest rate of attachment cells in regards to morphology. The result was statistically significant. However، the result for MTT and ALKAP index were not statistically significant.
Conclusion: In this study GBR-200 and TXT-200 membranes demonstrated better result in morphology changes of fibroblast cells in culture medium.
